Suppose that you digest the genomic DNA of a particular organism with Sau 3A (?GATC), where the arrow represents the cleavage site. Then you ligate the resulting fragments into a unique Bam HI (G?GATCC) cloning site of a plasmid vector. Would it be possible to isolate the cloned fragments from the vector using Bam HI? From what proportion of clones would it be possible?
